Physicochemical and microbial properties of dairy barn soils: a case study in Costa Rican farm-associated soils harboring the foodborne pathogen Listeria monocytogenes

Animal farming activities can influence soil properties that contribute to the survival
of foodborne pathogens like the ubiquitous Listeria monocytogenes. However, the presence of this
pathogen in farm-associated soils in Costa Rica has not been studied, which might provide new
insights regarding the environmental conditions associated with the establishment of this pathogen.
In this work, we correlated the presence of L. monocytogenes with the soil physicochemical properties
and bacterial community structure of soils associated with livestock activities, including a dairy barn
floor (DB) and a slaughterhouse holding pen (SH). A cropland (CL) was included as an example
of soil not associated with animal farming practices. We characterized the presence pattern of L.
monocytogenes via culture-dependent and culture-independent techniques (i.e., metabarcoding based
on 16S rRNA gene sequencing) and conducted a determination of physical, elemental and chemical
parameters with Fourier-transform infrared spectroscopy (FT-IR) to statistically determine the soil
properties that correlate with L. monocytogenes’ presence in the soil. L. monocytogenes was isolated from
DB samples and SH but not from CL. Subsequently, 16S rRNA gene-based metabarcoding showed
that the presence of L. monocytogenes was positively correlated with higher bacterial diversity, while
physicochemical analyses revealed that the total hydrogen and nitrogen contents of soil organic matter,
pH, and electrical conductivity were the main drivers of L. monocytogenes’ presence. Moreover, a CL
sample fertilized with animal-derived products showed DB-like physicochemical properties matching
conditions in favor of L. monocytogenes’ presence. Hence, our work emphasizes the significance of soil
as a primary source for the widespread dissemination of pathogens, particularly underscoring the
necessity for improved agricultural practices to prevent cross-contamination with L. monocytogenes.
Additionally, we highlight the importance of further understanding the biotic and abiotic factors in
facilitating the establishment of L. monocytogenes.

Tissue engineering aims to develop in-vitro substitutes of native tissues. One approach of
tissue engineering relies on using bioreactors combined with biomimetic scaffolds to produce study
models or in-vitro substitutes. Bioreactors provide control over environmental parameters, place
and hold a scaffold under desired characteristics, and apply mechanical stimulation to scaffolds.
Polymers are often used for fabricating tissue-engineering scaffolds. In this study, polycaprolactone
(PCL) collagen-coated microfilament scaffolds were cell-seeded with C2C12 myoblasts; then, these
were grown inside a custom-built bioreactor. Cell attachment and proliferation on the scaffolds were
investigated. A loading pattern was used for mechanical stimulation of the cell-seeded scaffolds.
Results showed that the microfilaments provided a suitable scaffold for myoblast anchorage and that
the custom-built bioreactor provided a qualified environment for the survival of the myoblasts on
the polymeric scaffold. This PCL-based microfilament scaffold located inside the bioreactor proved
to be a promising structure for the study of skeletal muscle models and can be used for mechanical
stimulation studies in tissue engineering applications.

para todos los grupos investigados.; In order to identify the relevant behavioral competencies for IT professionals and determine
if there is a gap between the competencies developed by academia and those required
by the job market in Costa Rica, this article analyses four stakeholder groups: literature, job
boards, the graduation profiles of IT programs in public universities, and the directors and
academic coordinators of these universities. The analysis identified 45 behavioral competencies,
of which only three coincided in their categorization as having a high or moderate
degree of importance. Notably, the analysis found no homogeneous competencies for all
the investigated groups.

aprender matemática y, en general, para desempeñarse en ambientes relacionados con esa disciplina.; The results of a study on the perception of mathematics as a male domain in official
day high school students in Costa Rica are presented. The study was conducted to evaluate the need
to develop intervention programs that promote that women have the same capabilities as men in
mathematics. “Male domain” is understood as the belief that men have better conditions than women
to learn mathematics and, in general, to perform in environments related to that discipline.
