Embriogénesis somática a partir de embriones inmaduros de arroz (Oryza sativa) subespecie indica: optimización de la regeneración de vitroplantas
Resumen
La presente investigación tuvo como objetivo optimizar la regeneración de
vitroplantas a partir de callos embriogénicos producidos de embriones inmaduros de
arroz (Oryza sativa. L var. CR-5272). Para ello, se recolectaron cariópsides en
estado de llenado del grano (leche media) de plantas cultivadas en invernadero. Se
realizó el procedimiento adaptado de Christou et al. (1991) y Christou & Ford (1995).
Se extrajeron 30 embriones inmaduros, se midieron con un vernier (Mitutoyo) y se
sometieron a un proceso de callogénesis en medio CC suplementado con 2 mg/L de
2,4-D y caseína hidrolizada (Potrykus et al., 1979). Se introdujo un total de 150
embriones (cinco repeticiones de 30 embriones cada una). Posteriormente, se evaluó
su regeneración a tres dosis de kinetina y ácido naftalenacético. Se utilizaron los
callos provenientes de 10 embriones por tratamiento. Se evaluó la cantidad de
vitroplantas obtenidas de cada embrión, así como la cantidad de callos necrosados u
oxidados. Se obtuvo un número variable de vitroplantas por embrión, desde cero
hasta 79 vitroplantas por embrión, con un promedio de 5,39 ± 12,06 vitroplantas por
embrión. La mayor cantidad de vitroplantas se obtuvo al utilizar embriones de entre
1,9 y 2 mm de longitud e inoculados en medio suplementado con 1mg/L de kinetina y
0,25 mg/L de ácido naftalenacético. The main objective of this investigation was to optimize the regeneration of plants from embriogenic calli induced from immature embryos of rice (Oryza sativa. L var. CR-5272). To accomplish this, immature seeds in filling state (medium milk) were collected from greenhouse-cultured plants. A procedure adapted from Christou et al. (1991) and Christou & Ford (1995) was performed. Thirty immature embryos were extracted, measured with a vernier (Mitutoyo) and callogenesis was induced by culture on CC media supplemented with 2 mg/L of 2,4-D and hydrolyzed casein (Potrykus et al., 1979). A total of 150 embryos were introduced (five repetitions of 30 embryos each). Later, their regeneration was evaluated at three concentrations of kinetin and naphtalenacetic acid. Calli obtained from ten embryos were inoculated on each treatment. The amount of plants obtained from each embryo was evaluated, along with the amount of oxidized and necrotized calli. A variable number of plants per embryo was obtained, from cero to 79 plants per embryo, with an average of 5,39 ± 12,06 plants per embryo. The largest amount of plants was achieved by using embryos between 1,9 and 2 mm long and inoculating them on CC media supplemented with 1mg/L kinetin and 0,25 mg/L naphtalenacetic acid.
